Streptobacillus moniliformis requires a number of conditions for successful cultivation in culture. Failure to provide all of the requirements can result in a false negative result.
Culture Condition
Requirement
atmosphere
microaerophilic with 8-10% CO2
nutritional supplementation of trypticase soy broth or agar
10-20% defibrinated blood, ascites fluid or animal serum
absence of inhibitors
no sodium polyanethol sulfonate
sufficient time
need to hold cultures since it may take up to 10 days to appear
appropriate temperature
37°C
If there is a strong clinical suspicion for infection with Streptobacillus moniliformis but cultures are negative, then polymerase chain reaction (PCR) should be used for diagnosis.
