Description

Griseofulvin is poorly soluble in water, which limits gastrointestinal absorption following an oral dose. Absorption can often be improved if certain steps are taken.

Intervention

poor drug dissolution secondary to particle size

ultramicronize the drug into tiny particles

poor absorption on an empty stomach

take with food, especially a fatty meal

poor water solubility

mixture with polyethylene glycol or preparation of micelles

poor absorption of a large dose

take a smaller dose more often (but this may make compliance a problem)

interference due to concurrent barbiturate therapy

avoid barbiturates while taking griseovulvin

 

Some patients are poor absorbers of griseofulvin despite all interventions.
